For blues, I usually keep it pretty simple. A clean or slightly edge-of-breakup amp, low to medium gain, bass around 4–5, mids 6–7, treble 5–6, and just a touch of spring reverb works well. If you have an overdrive, set the drive low and use it more as a boost than for heavy distortion. It also depends on what guitar and amp or modeler you're using, what gear do you have?
